Karl Barry Sharpless (lahir 28 April 1941) ialah seorang kimiawan Amerika Serikat yang dikenal dengan karyanya pada reaksi stereoselektif. Ia mendapat Penghargaan Nobel untuk bidang kimia tahun 2022 bersama Carolyn R. Bertozzi dan Morten P. Meldal atas hasil karya mereka untuk pengembangan kimia klik dan kimia bioorthogonal. [1]

Sharpless lahir di Philadelphia. Ia lulus dari Friends' Central School pada tahun 1959. Ia melanjutkan pendidikannya di Dartmouth College (1963) dan menerima Ph.D dari Stanford University pada tahun 1968. Ia melanjutkan karya pascadoktoral di Universitas Stanford dan Harvard. Ia memegang gelar kehormatan dari Universitas Teknik München.

Sharpless adalah profesor di Massachusetts Institute of Technology dan Stanford University. Sekarang ia memegang jabatan profesor W. M. Keck dalam kimia di The Scripps Research Institute.

Pada tahun 2001 ia memenangkan Nobel Kimia untuk karyanya pada reaksi oksidasi stereoselektif (epoksidasi Sharpless, dihidroksilasi asimetris Sharpless, oksiaminasi Sharpless). Hadiah ini dibagi dengan William S. Knowles dan Ryoji Noyori (untuk karya mereka pada hidrogenasi stereoselektif). Ia juga berhasil mengepoksidisasi (menggunakan asam tartarat rasemik) bola Fuckminster Fullerene C-86, menggunakan p-kresol sebagai pelarut. Sekarang ia menghabiskan sebagian besar waktunya dengan memperkenalkan kimia ketuk, seperangkat reaksi eksoterm yang amat selektif yang terjadi dalam keadaan sejuk; variannya yang paling berhasil adalah sikloadisi Huisgen azida alkina untuk membentuk 1,2,3-triazol.
