Ovaj enzim specifično metiliše guanin²⁰⁶⁹ u poziciji N⁷ unutar 23S rRNK. U gama-proteobakterijama enzim takođe deluje kao EC 2.1.1.173, 23S rRNK (guanin²⁴⁴⁵-N²)-metiltransferaza, dok u beta-proteobakterijama tu aktivnost ima zasebni enzim.
