Brian Tyree Henry lahir di Fayetteville, Carolina Utara, dan besar di Washington, D.C. Ayahnya merupakan seorang tentara dan ibunya merupakan seorang guru.^[1] Ia menempuh pendidikan di Morehouse College di Atlanta, Georgia sebagai pebisnis yang bertransisi menjadi aktor pada awal tahun 2000-an, lalu ia menerima gelar masternya dari Yale School of Drama di Universitas Yale.^[4]
