การทำงานโดยทั่วไปของไคเนส

โปรตีนไคเนส (protein kinase) เป็นเอนไซม์ไคเนสที่ทำหน้าที่ปรับแต่งโปรตีนอื่นโดยการเติมหมู่ฟอสเฟตในเชิงเคมี (ฟอสฟอริเลชั่น) กระบวนการฟอสฟอริเลชั่นมักส่งผลให้เกิดการเปลี่ยนแปลงเชิงหน้าที่ของโปรตีนเป้าหมาย (ซับสเตรท) ผ่านการเปลี่ยนแปลงการทำงานของเอนไซม์, ตำแหน่งเซลล์ หรือความเกี่ยวพันกับโปรตีนอื่น จีโนมของมนุษย์ประกอบด้วยยีนโปรตีนไคเนสประมาณ 500 ยีน คิดเป็น 2% ยีนมนุษย์ทั้งหมด[1]

กิจกรรมทางเคมีของโปรตีนไคเนสประกอบด้วยการกำจัดหมู่ฟอสเฟตจาก ATP และติดหมู่ฟอสเฟตด้วยพันธะโคเวเลนต์กับหนึ่งในสามกรดอะมิโนที่มีหมู่ไฮดรอกซิลอิสระ ไคเนสส่วนใหญ่ทำงานได้กับทั้งเซอรีน และ ธรีออนีน ไคเนสอื่น ๆ ทำงานกับไทรอซีน และยังมีอีกจำนวนหนึ่ง (ดูออล-สเพซิฟิซิที ไคเนส) ที่ทำงานได้กับทั้งสามตัว[2] นอกจากนี้ยังมีโปรตีนไคเนสอื่นที่สามารถฟอสฟอริเลตกรดอะมิโนอื่นได้ เช่น ฮิสทิดีน ไคเนสที่ฟอสฟอริเลตองค์ประกอบของฮิสทิดีน[3]
