Lappeenrannan kaupunginmuseon edeltäjä, Lappeenrannan Pallon kaupunginosaan Joutsenosta siirretty savupirttimuseo vihittiin käyttöön juhlavan Erkko-juhlan yhteydessä 24. kesäkuuta 1910.[1] Etelä-Saimaan Nuorisoseura piti museota yllä yhdeksän vuotta, minkä jälkeen museon ylläpitäminen siirtyi 18.7.1919 perustetulle Lappeenrannan kotiseutuyhdistykselle. Museo kärsi tilan puutteesta ja toimi myöhemmin Muinais-ja kansatieteellisenä museona Pekka Kinnusen talossa Snellmaninkatu 6:ssa sekä Kuninkaankadun (nykyinen Koulukatu) varrella sijainneen kirjastotalon yhteydessä. Savupirttimuseo siirrettiin Tyysterniemeen 1931.[2]

Lappeenrannan Kotiseutu- ja Museoyhdistys luovutti museon Lappeenrannan kaupungin haltuun 1945. Museo siirtyi Lappeenrannan Linnoituksen vartiorakennukseen, joka kunnostettiin museokäyttöön. Lappeenrannan kaupungin museo uusissa tiloissa vihittiin käyttöön 6. elokuuta 1949. Nykyisin tiloissa toimii Ratsuväkimuseo. Museo siirtyi nykyisiin tiloihinsa linnoituksen pohjoiskärjessä sijaitseviin kivimakasiineihin ja avautui Etelä-Karjalan maakuntamuseona 19. heinäkuuta 1962. Museotoiminta siirtyi samalla Etelä-Karjalan maakuntaliiton hallintaan. Etelä-Karjalan maakuntamuseon hoitajana toimi Johannes Hämäläinen vuosina 1962–1972.[3]
