Знаходзіцца на паўночны захад ад паўвострава Бутыя. З'яўляецца часткай тэрыторыі Канады. Складзены галоўным чынам вапнякамі. Плошча 33 339 км², востраў займае 40-ае месца па плошчы ў свеце і 10-ае ў Канадзе. Даўжыня берагавой лініі 2576 км[1]. Вышыня над узроўнем мора — да 320 м.

Востраў Прынца Уэльскага ўваходзіць у паўночнаамерыканскі экалагічны рэгіён Палярная тундра (англ.: Middle Arctic tundra), які вылучаецца Сусветным фондам дзікай прыроды.
