Odenkirk lahir di Naperville, Illinois . Dia adalah adik dari aktor Amerika dan komedian Bob Odenkirk, dan bekerja sebagai penulis, produser dan aktor di acara komedi sketsa komedi Mr Show dengan Bob dan David, yang menampilkan saudaranya sebagai lawan mainnya. Odenkirk melanjutkan untuk menulis untuk Tenacious D dan Futurama . Dia telah menulis dan eksekutif menghasilkan episode The Simpsons .
